Rapamycin (sirolimus)

The leading geroscience drug candidate is still off-label and unproven for healthy longevity.

Rapamycin inhibits mTOR, a nutrient-sensing pathway involved in growth, metabolism and autophagy. It extends lifespan in multiple mouse experiments, making it one of geroscience's most important drug candidates.

WHAT THE HUMAN DATA SAY

Evidence before enthusiasm.

Small trials in older or healthy adults suggest that selected low-dose or intermittent regimens can be feasible, but benefits across function, disease prevention and lifespan remain uncertain. Sirolimus is FDA-approved for other indications—not aging.
